Overview

Trastuzumab + capecitabine is a combination therapy used primarily in the treatment of HER2-positive breast cancer, especially in advanced or metastatic settings. Trastuzumab is a humanized monoclonal antibody that targets the extracellular domain of the human epidermal growth factor receptor 2 (HER2), inhibiting downstream signaling pathways involved in cell proliferation and survival. Capecitabine is an oral prodrug that is enzymatically converted to 5-fluorouracil (5-FU) within tumor cells, where it acts as an antimetabolite by inhibiting DNA synthesis and disrupting RNA function, leading to apoptosis. This combination exploits both targeted therapy against HER2 and cytotoxic chemotherapy for synergistic anti-tumor effects[5][6].
