Trastuzumab + docetaxel + platinum is a combination regimen used primarily in the treatment of HER2-positive breast cancer. Trastuzumab is a humanized monoclonal antibody that targets the extracellular domain of human epidermal growth factor receptor 2 (HER2), which is overexpressed in approximately 20% of breast cancers. Docetaxel is a taxane-class chemotherapeutic agent that inhibits microtubule depolymerization, leading to cell cycle arrest and apoptosis. Platinum agents (most commonly carboplatin or cisplatin) are DNA-damaging agents that form DNA crosslinks and inhibit DNA synthesis and function. The combination exploits synergistic effects between these drugs, resulting in high clinical activity for HER2-positive metastatic and early-stage breast cancer patients[1][5][6]. This regimen may also be referred to as TCH when carboplatin is used as the platinum component.
