Overview

Trastuzumab + doxorubicin is a combination therapy used primarily in the treatment of HER2-overexpressing breast cancer. Trastuzumab is a humanized monoclonal antibody that selectively binds to the extracellular domain of the human epidermal growth factor receptor 2 (HER2), inhibiting its dimerization and downstream signaling, which suppresses cancer cell growth and survival. Doxorubicin is an anthracycline antibiotic that intercalates DNA, inhibits topoisomerase II, and generates free radicals leading to cytotoxicity. The combination leverages trastuzumab’s targeted anti-HER2 activity with doxorubicin’s broad cytotoxic effects for synergistic efficacy in HER2-positive cancers[1][4][5][6].
