The trastuzumab + OKT3 bispecific antibody is a research-stage bispecific antibody (bsAb) construct designed as a T-cell engager (TCE). It incorporates the HER2-binding domains of trastuzumab and the CD3-binding domains of the OKT3 antibody (often in a humanized form). The mechanism involves simultaneously binding to HER2 on the surface of tumor cells and the CD3 complex on T cells, thereby bypassing the need for MHC-restricted antigen presentation and redirecting T-cell cytotoxic activity to kill HER2-positive cancer cells. Developed primarily as a platform technology by Absolute Antibody (a Vector Laboratories company), this construct has been used to evaluate the impact of different bispecific architectures—such as scFv placement and Fc engineering—on the stability, expression, and functional potency of multi-specific therapeutics.
