Overview

Trastuzumab + pertuzumab is a fixed-dose combination of two recombinant humanized monoclonal antibodies targeting the human epidermal growth factor receptor 2 (HER2). Both components are produced using Chinese hamster ovary (CHO) cells. Trastuzumab binds to subdomain IV of the HER2 extracellular domain, inhibiting ligand-independent HER2 signaling and inducing antibody-dependent cellular cytotoxicity (ADCC). Pertuzumab binds to subdomain II of HER2, blocking ligand-dependent heterodimerization with other HER family members and further inhibiting downstream signaling. The combination provides complementary mechanisms that result in more comprehensive blockade of the HER2 pathway and enhanced antitumor activity compared to either agent alone. This dual-targeted therapy is primarily indicated for early-stage and metastatic HER2-positive breast cancer in combination with chemotherapy[1][3][6].
