Trastuzumab + pyrotinib is a combination therapy used primarily for the treatment of HER2-positive breast cancer, including metastatic and early or locally advanced disease. Trastuzumab is a monoclonal antibody that targets the extracellular domain of human epidermal growth factor receptor 2 (HER2), inhibiting HER2-mediated signaling and inducing antibody-dependent cell-mediated cytotoxicity. Pyrotinib is an oral, irreversible tyrosine kinase inhibitor (TKI) that targets multiple members of the HER family, including epidermal growth factor receptor (EGFR/HER1), HER2, and HER4. The combination provides dual blockade of the HER2 pathway by simultaneously targeting both extracellular and intracellular domains, resulting in synergistic inhibition of tumor cell proliferation and survival. This regimen has demonstrated improved progression-free survival in clinical trials for patients with untreated or previously treated HER2-positive metastatic breast cancer[1][2][3][5][6].
