This combination therapeutic regimen is being evaluated in the T-sunflower Phase II clinical trial led by Fudan University for the treatment of ER+/HER2+ advanced breast cancer. The regimen integrates multiple targeted approaches: trastuzumab, a monoclonal antibody that binds to the extracellular domain of HER2; pyrotinib, a small molecule tyrosine kinase inhibitor targeting HER2 and EGFR; dalpiciclib, a selective inhibitor of CDK4 and CDK6; and fulvestrant, an estrogen receptor antagonist. This multi-pronged strategy aims to simultaneously inhibit the HER2 signaling pathway, block cell cycle progression, and suppress hormone-driven growth to overcome resistance and improve clinical outcomes in patients with hormone receptor-positive, HER2-positive metastatic breast cancer.
