Overview

Trastuzumab + serplulimab is a combination therapy consisting of the HER2-targeted monoclonal antibody trastuzumab and the PD-1-targeted monoclonal antibody serplulimab. This combination is being investigated, often alongside chemotherapy, for the treatment of HER2-overexpressing advanced malignancies, specifically biliary tract cancer and urothelial carcinoma. Trastuzumab works by binding to the HER2 receptor on tumor cells, inhibiting proliferation and promoting antibody-dependent cellular cytotoxicity (ADCC). Serplulimab (HLX10) is a humanized IgG4 monoclonal antibody that blocks the PD-1/PD-L1 pathway, thereby reactivating the immune system's anti-tumor response. The combination is currently being evaluated in the Phase II SHIELD trial (NCT05501444) led by investigator Xiujuan Qu for patients who have failed standard therapies.
