Overview

trastuzumab + ST-PHPMA is an antibody-polymer conjugate consisting of the HER2-targeting monoclonal antibody trastuzumab covalently conjugated to a semitelechelic poly[N-(2-hydroxypropyl)methacrylamide] (ST-PHPMA) copolymer backbone. Developed by researchers at the University of Maryland, this macromolecular drug delivery system is designed to actively target HER2-overexpressing cancer cells, such as those in HER2-positive breast cancer. By utilizing a semitelechelic polymer structure, the conjugate allows for single-point attachment to the antibody, avoiding cross-linking and maintaining the antibody's bioactivity. In preclinical studies, the conjugate demonstrated prolonged inhibitory effects on the MAPK and Akt signaling pathways compared to free trastuzumab, suggesting that polymer conjugation can extend the therapeutic activity of the antibody while retaining its efficacy.
