Overview

**Trastuzumab deruxtecan + bevacizumab** is an investigational combination therapy for cancer, notably HER2-expressing ovarian cancer and potentially other HER2-positive or HER2-low solid tumors. Trastuzumab deruxtecan is an antibody-drug conjugate targeting HER2 (human epidermal growth factor receptor 2), combining a humanized monoclonal antibody with a cytotoxic payload (deruxtecan, a topoisomerase I inhibitor) delivered via a cleavable peptide linker. Bevacizumab is a humanized monoclonal antibody targeting VEGF-A (vascular endothelial growth factor A), inhibiting angiogenesis. Together, this regimen exploits targeted cell killing, immunomodulation, and anti-angiogenic mechanisms to enhance antitumor effect. The regimen is currently being studied in phase 3 trials for HER2-expressing ovarian cancer and has preclinical and clinical rationale for synergistic use in breast and other cancers[1][4][2][5][6][8].
