Overview

A combination of three anti-cancer agents: **trastuzumab deruxtecan (T-DXd)**, an antibody-drug conjugate targeting HER2; **durvalumab**, a monoclonal antibody checkpoint inhibitor targeting PD-L1; and **carboplatin**, a platinum-based chemotherapy agent. This triplet combination is under investigation for use as first-line treatment in patients with advanced or metastatic non-squamous non-small cell lung cancer (NSCLC) with HER2 overexpression. Trastuzumab deruxtecan binds HER2-expressing cancer cells, delivering a topoisomerase I inhibitor payload, causing targeted DNA damage and cell death. Durvalumab blocks PD-L1, enhancing anti-tumor immune response. Carboplatin forms DNA crosslinks via alkylation, inhibiting cell replication and leading to apoptosis[1][3][5][6].
