Overview

This is a combination therapy of three agents: - **Trastuzumab deruxtecan (T-DXd, DS-8201a, Enhertu)**: an antibody-drug conjugate composed of a humanized anti-HER2 monoclonal antibody linked to a topoisomerase I inhibitor cytotoxic payload. It delivers chemotherapy directly to HER2-overexpressing tumor cells, causing DNA damage and apoptosis. - **Rilvegostomig (AZD2936)**: a novel bispecific monoclonal antibody targeting both PD-1 and TIGIT immune checkpoints. It is designed to enhance antitumor immune response by dual blockade, increasing T-cell mediated tumor cell killing. - **Carboplatin**: a platinum-based chemotherapy agent that induces DNA crosslinking and apoptosis in rapidly dividing cells. This combination is being studied as a first-line therapy for HER2-expressing, mismatch repair proficient, advanced or recurrent endometrial cancer in Phase 3 clinical trials. It is administered intravenously.
