Overview
AD816 is a fixed-dose oral combination of trazodone and viloxazine in development for the treatment of obstructive sleep apnea. Trazodone is a serotonin receptor antagonist and reuptake inhibitor, while viloxazine is a serotonin-norepinephrine modulating agent. This combination is intended to harness complementary effects on neurochemical pathways involved in sleep regulation and upper airway tone. The drug is being developed by Apnimed, with the original invention attributed to Mass General Brigham. Its mechanisms include antagonism at serotonin 5-HT2A and 5-HT2C receptors, inhibition of serotonin and adrenergic reuptake, and alpha adrenergic receptor agonism. Phase II studies have been completed in the USA for obstructive sleep apnea.
