Overview

A combination immunotherapy consisting of the anti-CTLA-4 monoclonal antibody tremelimumab and recombinant interferon alfa-2b investigated primarily for advanced melanoma. Tremelimumab blocks CTLA-4 to enhance T-cell activation and antitumor immunity, while interferon alfa-2b exerts immunomodulatory and antineoplastic effects; together they were studied for potential additive or synergistic activity. A phase II study administered tremelimumab 15 mg/kg IV every 12 weeks with concurrent high-dose interferon alfa-2b induction (20 MU/m2/day IV, 5 days/week for 4 weeks) followed by maintenance (10 MU/m2/day SC three times weekly for 8 weeks), showing acceptable toxicity and signs of durable antitumor efficacy in metastatic melanoma patients.[4] Tremelimumab is an IgG2 monoclonal antibody targeting CTLA-4, developed by AstraZeneca/MedImmune; interferon alfa-2b is a recombinant type I interferon historically marketed as Intron A.[2][4][6]
