Overview

**Tremelimumab + nivolumab** is a combination of two monoclonal antibody immune checkpoint inhibitors used in oncology. Tremelimumab is a fully human IgG2 monoclonal antibody targeting cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), acting as a negative regulator of activated T cells and thus enabling increased anti-tumor T cell responses. Nivolumab is an IgG4 monoclonal antibody targeting programmed death-1 (PD-1) receptor, blocking its interaction with ligands PD-L1 and PD-L2, thereby relieving immunosuppression of T cells in the tumor microenvironment. The combination works via complementary mechanisms to boost T-cell-mediated cancer cell destruction. Primary indications include unresectable hepatocellular carcinoma and other advanced solid tumors, with clinical trials demonstrating improved survival and response rates over monotherapies and previous standards of care[6].
