Twyneo is a topical fixed-dose combination cream containing microencapsulated tretinoin (0.1%) and microencapsulated benzoyl peroxide (3%). Developed by Sol-Gel Technologies using its proprietary silica-based microencapsulation technology, it is the first FDA-approved product to combine these two ingredients, which are typically chemically incompatible because benzoyl peroxide causes the oxidative degradation of tretinoin. The microencapsulation technology separately encapsulates each active ingredient in silica core shells, protecting the tretinoin from degradation and allowing for a controlled, gradual release onto the skin to improve efficacy and reduce irritation. Tretinoin is a retinoid that acts as an agonist for retinoic acid receptors (RARα, RARβ, and RARγ), modulating gene expression to normalize follicular desquamation and reduce the formation of comedones. Benzoyl peroxide provides complementary antibacterial activity against *Cutibacterium acnes* and possesses keratolytic properties. Twyneo is indicated for the treatment of inflammatory and noninflammatory lesions of acne vulgaris in adults and pediatric patients aged 9 years and older.
