Overview

This is a **combination drug** composed of triciribine phosphate monohydrate (also called PTX-200 or TCN-PM) and cytarabine, developed for the treatment of relapsed or refractory acute myeloid leukemia (AML). **Triciribine phosphate monohydrate** is a small molecule inhibitor of all three isoforms of the serine/threonine kinase AKT. Inhibition of AKT, which is constitutively activated in many malignancies including AML, disrupts cell survival and proliferation pathways. **Cytarabine** is a nucleoside analog with antimetabolite activity, interfering with DNA synthesis and inducing cytotoxicity in rapidly dividing hematologic cancer cells. Combination therapy is being investigated based on the rationale that AKT inhibition may enhance the cytotoxic effects of cytarabine and overcome resistance mechanisms. The combination is being developed and tested by Prescient Therapeutics in Phase I/II clinical trials for AML, with orphan drug designation granted by FDA for this indication[3][5][7].
