Overview

Trifluridine + tipiracil + bevacizumab is a combination therapy used for the treatment of metastatic colorectal cancer in patients who have previously received other chemotherapy regimens. Trifluridine is a thymidine-based nucleoside analogue that incorporates into DNA, causing dysfunction and cell death. Tipiracil inhibits thymidine phosphorylase, thereby increasing the bioavailability of trifluridine by preventing its breakdown. Bevacizumab is a monoclonal antibody that targets vascular endothelial growth factor (VEGF), inhibiting angiogenesis and thus starving tumors of oxygen and nutrients. This combination has demonstrated improved survival outcomes in clinical trials for refractory metastatic colorectal cancer[1][2][5][6][8].
