Drug intelligence / Profile preview
**trilaciclib + sacituzumab govitecan-hziy** is a combination regimen being investigated for the treatment of unresectable locally advanced or metastatic triple-negative breast cancer (TNBC). **Trilaciclib** is a cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor that protects hematopoietic stem and progenitor cells (HSPCs) from chemotherapy-induced myelosuppression by transiently arresting them in the G1 phase of the cell cycle. **Sacituzumab govitecan-hziy** is an antibody-drug conjugate (ADC) that targets trophoblast cell-surface antigen 2 (Trop-2) and delivers the cytotoxic payload SN-38 (an active metabolite of irinotecan). Administering trilaciclib before sacituzumab govitecan-hziy aims to reduce the rate and severity of adverse events, notably myelosuppression and gastrointestinal toxicity, without compromising anti-tumor efficacy. The regimen is in phase 2 clinical trials and has shown promising reductions in severe hematological and non-hematological toxicities compared to the ADC alone, potentially enhancing patient safety and treatment continuity[1][2][3][4].
