TriMix is an mRNA-based immunostimulatory drug substance consisting of a 1:1:1 mixture of three mRNA sequences encoding CD40 ligand (CD40L), CD70, and a constitutively active form of Toll-like receptor 4 (caTLR4). It is designed to function as a potent genetic adjuvant by targeting and activating dendritic cells (DCs) and other antigen-presenting cells in vivo. By providing three critical signals—CD40 ligation for DC maturation, CD70 for T-cell costimulation via CD27, and TLR4 signaling for innate activation—TriMix enhances the induction of robust, antigen-specific T-cell responses. Within the iHIVARNA-01 program, TriMix is combined with the HIVACAT T-cell immunogen (HTI) mRNA to form a therapeutic vaccine for chronic HIV-1 infection, aiming to achieve a functional cure by controlling viral replication without antiretroviral therapy. The drug is administered via intranodal injection and has been evaluated in Phase I and Phase IIa clinical trials.
