TriMix-DC + ipilimumab is an investigational combination immunotherapy for the treatment of advanced malignant melanoma. It consists of an autologous dendritic cell-based vaccine (TriMix-DC) and the immune checkpoint inhibitor ipilimumab. The TriMix-DC component involves monocyte-derived dendritic cells electroporated with synthetic mRNA encoding a 'TriMix' of activating molecules (CD70, CD40 ligand, and constitutively active TLR4) and four melanoma-associated antigens (tyrosinase, gp100, MAGE-A3, and MAGE-C2). This vaccine is designed to stimulate a robust T-cell response against tumor cells. Ipilimumab, a monoclonal antibody targeting CTLA-4, is added to enhance this immune response by removing inhibitory signals on T-cells. The combination has been studied in Phase II clinical trials led by researchers at Universitair Ziekenhuis Brussel.
