Overview

This is a **combination product** consisting of a trivalent inactivated influenza vaccine (TIV)—commonly a split virion vaccine containing inactivated influenza A (H1N1 and H3N2) and B virus components—and **JVRS-100**, a cationic liposome–DNA complex adjuvant[5][1][3]. JVRS-100 acts as a potent immunostimulant, enhancing both humoral (antibody) and cell-mediated (CD4⁺ and CD8⁺ T cell) immune responses to influenza antigens[1][5][3]. Animal and Phase 1 clinical studies showed that the combination increases hemagglutination inhibition (HAI) and neutralizing antibody titers, as well as robust T cell responses, compared to the vaccine alone[5][1][3]. This approach aims to improve the efficacy of influenza vaccination, especially in populations with weaker immune responses such as the elderly, and potentially enable lower antigen dose usage or improved efficacy against drifted viral strains[1][5]. The main mechanism is enhancement of the immune response through adjuvant activity; JVRS-100's unique cationic lipid/DNA structure generates strong Th1 and cytotoxic T cell (CD8⁺) responses, not seen with the split influenza vaccine alone[5][3].
