Overview

**Trivirus-specific cytotoxic T-lymphocytes** are an investigational, donor-derived adoptive T-cell immunotherapy developed at Baylor College of Medicine for persistent or recurrent **Epstein-Barr virus, cytomegalovirus, and adenovirus** infection after allogeneic hematopoietic stem cell transplantation. The product consists of ex vivo expanded cytotoxic T cells with antiviral specificity against EBV, CMV, and adenovirus antigens. A rapid manufacturing approach uses dendritic cells nucleofected with DNA plasmids encoding EBV LMP2, EBNA1, and BZLF1; adenovirus Hexon and Penton; and CMV pp65 and IE1, generating a polyclonal antiviral T-cell product. After intravenous administration, the HLA-matched donor-derived T cells recognize viral peptide-HLA complexes on infected cells and mediate cytotoxic clearance of infected cells, restoring antiviral cellular immunity in immunocompromised transplant recipients.
