TRK-950 + FOLFIRI is a combination therapy under investigation for the treatment of advanced solid tumors, including metastatic colorectal cancer. TRK-950 is a first-in-class, humanized monoclonal antibody (IgG1) that targets cytoplasmic activation/proliferation-associated protein-1 (CAPRIN-1), which is highly expressed in many solid tumor types but largely absent from normal tissues. TRK-950's mechanism involves antibody-dependent cell-mediated cytotoxicity and antibody-dependent cell-mediated phagocytosis, leading to tumor cell death. FOLFIRI is a well-established chemotherapy regimen consisting of folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan, which work together to disrupt DNA synthesis and replication, thereby slowing tumor growth and inducing cancer cell death. The combination aims to leverage TRK-950's immunologic killing effect and FOLFIRI's cytotoxicity for potentially synergistic anti-tumor activity. The combination is currently being studied in phase Ib and II trials for advanced cancers, primarily those with CAPRIN-1 expression[1][3][5].
