Overview

TRP-2:180-188 peptide is a synthetic peptide vaccine and immunotherapeutic agent derived from the tyrosinase-related protein 2 (TRP-2) enzyme, also known as dopachrome tautomerase. TRP-2 is a melanosomal membrane protein involved in the synthesis of melanin and is highly expressed in melanoma cells, making it an ideal tumor-associated antigen. The specific fragment spanning amino acids 180 to 188 (sequence: SVYDFFVWL) is a highly immunogenic epitope restricted by HLA-A*02:01 in humans and H-2Kb in mice. When administered, typically in combination with adjuvants such as Montanide ISA-51 or loaded onto dendritic cells, the peptide is presented by major histocompatibility complex (MHC) class I molecules to CD8+ T cells. This interaction stimulates the expansion of antigen-specific cytotoxic T lymphocytes (CTLs) that can recognize and lyse TRP-2-expressing tumor cells. This peptide has been primarily investigated in clinical trials for the treatment of malignant melanoma, often as part of multi-peptide vaccine cocktails or in combination with immune checkpoint inhibitors.
