Overview

Trumenba (bivalent rLP2086) is a recombinant protein vaccine developed by Pfizer for the prevention of invasive meningococcal disease caused by Neisseria meningitidis serogroup B. The vaccine is composed of two lipidated factor H-binding protein (fHBP) variants, one from subfamily A (A05) and one from subfamily B (B01), which are recombinantly expressed in Escherichia coli. fHBP is a surface-exposed lipoprotein that enables the bacteria to evade host immune defenses by binding human factor H. By inducing a robust bactericidal antibody response against these proteins, Trumenba facilitates complement-mediated killing of the bacteria. The specific designation 'rLP2086 + saline' refers to a clinical trial intervention (such as in NCT01352845) where the vaccine is administered alongside saline injections, which serve as placebos for other vaccines in studies assessing concomitant administration. Trumenba was the first serogroup B meningococcal vaccine approved in the United States, receiving accelerated approval in 2014 for use in individuals aged 10 through 25 years.
