Overview

TRX518 + gemcitabine is an investigational combination therapy for cancer. TRX518 is a humanized monoclonal antibody that targets the glucocorticoid-induced TNF receptor–related protein (GITR) on T cells. By agonizing GITR, TRX518 enhances immune responses against tumors by activating effector T cells and reducing immunosuppressive regulatory T cells within the tumor microenvironment[1][2][4][5]. Gemcitabine is a nucleoside analog chemotherapeutic agent that inhibits DNA synthesis by incorporating into DNA during replication, leading to cell cycle arrest and apoptosis in rapidly dividing cancer cells[6][8]. The combination has been studied in early-phase clinical trials for advanced solid tumors to assess safety and preliminary efficacy[2].
