Tryptophan + vitamin B6 is a combination of the essential amino acid L-tryptophan and pyridoxine (vitamin B6). L-tryptophan serves as the primary metabolic precursor for the biosynthesis of the neurotransmitter serotonin (5-hydroxytryptamine) and the hormone melatonin. The conversion of tryptophan to serotonin involves two steps: the rate-limiting hydroxylation to 5-hydroxytryptophan (5-HTP) by tryptophan hydroxylase, followed by decarboxylation to serotonin by the enzyme aromatic L-amino acid decarboxylase (AAAD). Vitamin B6, in its active form pyridoxal 5'-phosphate (PLP), is a critical cofactor for AAAD. This combination is primarily used as a dietary supplement or pharmaceutical intervention to support mood regulation, improve sleep quality, and manage symptoms of depression or anxiety by optimizing endogenous serotonin synthesis. Historically, L-tryptophan faced regulatory scrutiny in the late 1980s due to contamination-related health issues, but it has since been reintroduced under stringent manufacturing standards.
