TSC-203-A0201 + TSC-204-A0101 is a combination of two investigational autologous, engineered T cell receptor (TCR)-T cell therapies developed by TScan Therapeutics for the treatment of solid tumors. Each product targets a specific cancer-associated antigen presented on distinct HLA types: * **TSC-203-A0201** targets PRAME (PReferentially expressed Antigen in Melanoma) presented on HLA-A*02:01. * **TSC-204-A0101** targets MAGEA1 (Melanoma Antigen Gene A1) presented on HLA-A*01:01. These therapies are part of the "T-Plex" multiplex platform, designed to address tumor heterogeneity and overcome immune evasion by targeting multiple antigens and HLA types simultaneously. The products are enhanced with co-delivery of CD8α/β to engage helper T cells and may include a dominant-negative transforming growth factor beta receptor II (DN-TGFβRII) to improve expansion and persistence of the infused cells[2][4][5][8]. Both agents are currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
