Overview

TSC-204-A0201 + TSC-203-A0201 is a combination of two autologous, engineered T cell receptor (TCR)-T cell therapies developed by TScan Therapeutics for the treatment of solid tumors. Each component targets a different tumor-associated antigen presented on HLA type A*02:01. Specifically, TSC-204-A0201 targets Melanoma-associated antigen 1 (MAGE-A1) and TSC-203-A0201 targets Preferentially expressed antigen in melanoma (PRAME). Both are part of the "T-Plex" multiplexed solid tumor program designed to address tumor heterogeneity and HLA loss by enabling customized combinations of multiple antigen-specific engineered T cells for each patient. The therapy involves leukapheresis to collect patient cells, which are then genetically modified ex vivo to express specific high-affinity cancer-targeting receptors before being reinfused into the patient following lymphodepletion[2][3][5][6][8].
