TSC-204-A0201 + TSC-204-A0101 is a combination of two autologous, engineered T cell receptor (TCR)-T cell therapies developed by TScan Therapeutics. Each product consists of patient-derived (autologous) T cells genetically modified to express a specific high-affinity, naturally occurring TCR targeting the cancer-testis antigen MAGE-A1 presented on different HLA types—HLA-A*02:01 for TSC-204-A0201 and HLA-A*01:01 for TSC-204-A0101. These therapies are designed to recognize and kill tumor cells expressing MAGE-A1 while sparing normal tissues, as MAGE-A1 is highly expressed in solid tumors but largely absent from healthy tissues except testis. The engineered cells also co-express CD8α/β coreceptors to enhance helper and cytotoxic function, as well as a dominant-negative form of the transforming growth factor beta receptor II (DN-TGFβRII) to improve persistence and resistance to immunosuppression within the tumor microenvironment. This combination is being evaluated in early-phase clinical trials for patients with locally advanced or metastatic solid tumors that are positive for both MAGE‑A1 antigen and the relevant HLA type[2][5][6].
