TSC-204-A0201 + TSC-204-C0702 is a combination of two investigational, autologous, engineered T cell receptor (TCR)-T cell therapies developed by TScan Therapeutics. Each product targets the melanoma-associated antigen 1 (MAGE-A1), but is restricted to different human leukocyte antigen (HLA) types: HLA-A*02:01 for TSC-204-A0201 and HLA-C*07:02 for TSC-204-C0702. These therapies are designed to recognize and kill tumor cells expressing MAGE-A1 in patients whose tumors present the relevant HLA type. The combination is part of a multiplexed approach ("T-Plex") that aims to overcome tumor heterogeneity and HLA loss by administering multiple customized TCR-T products simultaneously or sequentially, based on each patient’s tumor profile. The primary indications under investigation include solid tumors such as head and neck cancer, melanoma, cervical cancer, and non-small cell lung cancer[3][4][5][7].
