Overview

TSEBT + pembrolizumab is a combination therapy consisting of total skin electron beam therapy (TSEBT), a specialized form of external beam radiation targeting the entire skin surface, and pembrolizumab, a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1). This regimen is being investigated primarily for the treatment of advanced or relapsed cutaneous T-cell lymphoma (CTCL), including mycosis fungoides and Sézary syndrome. The rationale for this combination is that TSEBT rapidly reduces tumor burden in the skin while subsequent or concurrent administration of pembrolizumab may enhance systemic anti-tumor immunity by blocking PD-1-mediated inhibition of T cells. Early-phase clinical trials are assessing safety, tolerability, and preliminary efficacy in these patient populations[1][3].
