The molecule is engineered to simultaneously target the Poliovirus receptor (PVR, also known as CD155) on the surface of tumor cells and the CD3 receptor on T cells. By co-engaging these two targets, TSRF 786C acts as a T-cell engager, facilitating the formation of an immunological synapse between cytotoxic T cells and malignant cells. This interaction triggers T-cell activation and the subsequent release of perforins and granzymes, leading to the targeted lysis of PVR-expressing tumor cells. PVR is a known immune checkpoint ligand that is frequently upregulated in solid tumors and hematologic malignancies, making it a strategic target for redirecting immune responses and overcoming tumor-mediated immunosuppression.
