TST001 + nivolumab + mFOLFOX6 is an investigational combination therapy for advanced gastric and gastroesophageal junction (G/GEJ) cancers. It consists of: - **TST001 (osemitamab):** a humanized monoclonal antibody targeting claudin 18.2 (CLDN18.2), designed to kill cancer cells via antibody-dependent cellular cytotoxicity (ADCC) and possibly other immune mechanisms. - **Nivolumab:** an immune checkpoint inhibitor (anti-PD-1 antibody) that enhances T-cell mediated anti-tumor responses. - **mFOLFOX6:** a chemotherapy regimen containing oxaliplatin (a platinum-based chemotherapy), leucovorin (folinic acid), and fluorouracil (5-FU), used as a backbone therapy in various GI cancers. The triple regimen is being tested primarily as a first-line therapy for HER2-negative or unknown, unresectable, locally advanced, or metastatic gastric or gastroesophageal junction cancer, regardless of CLDN18.2 or PD-L1 expression. Early-phase studies show promising objective response and survival benefit, particularly in patients with higher CLDN18.2 expression[3][5].
