TTI-101 + pembrolizumab is an investigational combination therapy consisting of **TTI-101**, a first-in-class, orally administered small molecule inhibitor of **signal transducer and activator of transcription 3 (STAT3)**, and **pembrolizumab**, a monoclonal antibody and immune checkpoint inhibitor targeting **programmed cell death protein 1 (PD-1)**. The combination is being studied for safety, tolerability, and preliminary efficacy in patients with locally advanced or metastatic, and unresectable **hepatocellular carcinoma (HCC)**, and is also under investigation for recurrent or metastatic head and neck squamous cell carcinoma (HNSCC). TTI-101 inhibits STAT3, which is involved in tumor cell survival, proliferation, immune evasion, and is frequently activated in HCC. Pembrolizumab blocks PD-1, enhancing T-cell mediated immune response against tumor cells. The rationale for the combination is that dual inhibition of STAT3 signaling and the PD-1 pathway may enhance antitumor activity and overcome resistance to checkpoint blockade[1][3][6][7].
