TTRNA-DC + TTRNA-xALT is an investigational autologous cellular immunotherapy developed by the University of Florida for the treatment of newly diagnosed diffuse intrinsic pontine glioma (DIPG) and other brain stem gliomas. The treatment regimen, evaluated in the Phase I BRAVO trial, involves a multi-modal approach: first, a dendritic cell vaccine is loaded with total tumor messenger RNA (TTRNA) extracted from the patient's own tumor to prime the immune system against a comprehensive set of tumor-specific antigens. Second, the protocol includes the adoptive transfer of ex vivo expanded autologous tumor-reactive lymphocytes (TTRNA-xALT) to provide a robust effector T-cell population. This immunotherapy is typically administered following focal radiotherapy and may be combined with lymphodepleting chemotherapy (temozolomide) and hematopoietic stem cell support to optimize the expansion and anti-tumor activity of the transferred cells.
