This is a combination regimen consisting of TTX-030 (an anti-CD39 monoclonal antibody), budigalimab (an anti-PD-1 monoclonal antibody), nab-paclitaxel (a nanoparticle albumin-bound formulation of paclitaxel), and gemcitabine (a nucleoside analog chemotherapy agent). - **TTX-030** targets CD39, an enzyme involved in the immunosuppressive conversion of extracellular ATP to adenosine in the tumor microenvironment, thereby aiming to enhance anti-tumor immune responses by reducing adenosine-mediated immune suppression[1][3][5]. - **Budigalimab** is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), promoting T-cell mediated immune activity against tumors[2][4]. - **Nab-paclitaxel** (albumin-bound paclitaxel) inhibits microtubule disassembly, disrupting mitosis; it is commonly used as a chemotherapeutic agent in various solid tumors. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis, leading to apoptosis, and is widely used in the treatment of pancreatic and other solid tumors. This combination has been studied primarily in metastatic pancreatic ductal adenocarcinoma, where early clinical data demonstrate tolerability and promising efficacy outcomes[1][5]. TTX-030 is being developed by Trishula Therapeutics in collaboration with AbbVie[1][5].
