TTX-080 + cetuximab

TTX-080 + cetuximab is a combination of TTX-080, a novel, first-in-class monoclonal antibody targeting human leukocyte antigen-G (HLA-G), and cetuximab, an epidermal growth factor receptor (EGFR) inhibitor monoclonal antibody. TTX-080 is designed to block HLA-G, an immunosuppressive MHC class I molecule expressed by tumors to evade immune destruction, thereby reactivating anti-tumor immunity. Cetuximab binds to and inhibits EGFR, blocking downstream signaling pathways that promote tumor cell proliferation and survival. The combination is under clinical evaluation for biomarker-defined metastatic colorectal cancer (mCRC) and head and neck squamous cell carcinoma (HNSCC), with evidence of promising clinical activity, particularly in biomarker-defined, anti-EGFR-naïve patients who have received multiple prior therapies. TTX-080 induces immune activation both peripherally and within the tumor microenvironment and may synergize with cetuximab’s antitumor effects by further reducing immune escape and potentiating cytotoxic effects[2][5][1].
