TUB-040 + carboplatin + bevacizumab is an investigational combination regimen being evaluated for the treatment of platinum-sensitive high-grade epithelial serous or endometrioid ovarian cancer. TUB-040 is a next-generation antibody-drug conjugate (ADC) targeting NaPi2b (SLC34A2), a sodium-dependent phosphate transporter overexpressed in ovarian and lung cancers. It utilizes Tubulis' proprietary Tubutecan/P5 cleavable linker to attach the topoisomerase-I inhibitor payload, exatecan, with a drug-to-antibody ratio (DAR) of 8. Carboplatin is a platinum-based alkylating agent that interferes with DNA repair, while bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor (VEGF) to inhibit angiogenesis. This triplet combination aims to improve outcomes in earlier lines of ovarian cancer therapy by combining targeted cytotoxic delivery with systemic chemotherapy and anti-angiogenic effects.
