Overview

This regimen is a combination of five anticancer drugs: - **Tucatinib** is an oral, highly selective small molecule inhibitor of the human epidermal growth factor receptor 2 (HER2) tyrosine kinase, used primarily for HER2-positive cancers[1][3]. - **Trastuzumab** is a monoclonal antibody targeting the extracellular domain of HER2, inhibiting its signaling and inducing antibody-dependent cellular cytotoxicity (ADCC). - **Pembrolizumab** is a monoclonal antibody that inhibits programmed cell death protein 1 (PD-1), promoting immune activation and antitumor responses. - **Capecitabine** is an oral prodrug converted to 5-fluorouracil (5-FU), a pyrimidine antimetabolite that inhibits DNA synthesis. - **Oxaliplatin** is a platinum-based chemotherapy that cross-links DNA, leading to apoptosis. This combination acts through both targeted inhibition of HER2 signaling, immune activation via checkpoint inhibition, and cytotoxic chemotherapy. It is being investigated mainly for advanced or metastatic HER2-positive cancers, including HER2-positive breast cancer and potential evaluation in other HER2-positive solid tumors[1][3][5].
