This is an autologous cellular immunotherapy developed by Herlev Hospital for the treatment of malignant melanoma. The therapy involves harvesting a patient's own dendritic cells and loading (pulsing) them with specific tumor antigens to serve as professional antigen-presenting cells. For HLA-A2 positive patients, the dendritic cells are pulsed with a cocktail of peptides derived from p53, survivin, and telomerase. For HLA-A2 negative patients, the cells are pulsed with allogeneic tumor lysate. Once re-administered to the patient via intradermal injection, these matured dendritic cells are intended to prime and activate a T-cell mediated immune response against the tumor. In clinical protocols, this vaccine is typically administered as part of a multi-modal regimen including low-dose cyclophosphamide to deplete regulatory T cells, celecoxib to inhibit COX-2, and interleukin-2 to support the expansion of the induced T-cell population.
