Overview

Tumor-draining lymph node-derived lymphocytes (LNL) is an autologous adoptive cell therapy being developed by Sun Yat-Sen Memorial Hospital. Unlike traditional tumor-infiltrating lymphocytes (TILs) which are often harvested from the tumor microenvironment in an exhausted state, LNLs are isolated from the patient's sentinel lymph nodes. These nodes serve as a reservoir for tumor-reactive T cells that have been primed by tumor antigens but have not yet undergone the terminal exhaustion associated with the immunosuppressive tumor milieu. The lymphocytes are isolated via sentinel lymph node biopsy (SLNB), expanded *in vitro*, and re-infused into the patient following lymphodepleting chemotherapy. This approach aims to provide a robust population of effector T cells capable of targeting and eliminating residual or resistant cancer cells, particularly in patients with HER2-negative breast cancer who show a poor response to standard neoadjuvant chemotherapy.
