Overview

The tumor lysate-pulsed dendritic cell vaccine developed by the University of Michigan Rogel Cancer Center is an autologous cellular immunotherapy investigated for the treatment of high-risk pediatric solid tumors, including sarcoma, neuroblastoma, and Wilms' tumor. The vaccine is prepared by harvesting a patient's own dendritic cells and loading them with a lysate derived from the patient's own tumor tissue, which contains a broad array of tumor-associated antigens. When administered back to the patient—typically in the setting of immune reconstitution following autologous hematopoietic stem cell transplantation—these dendritic cells present the tumor antigens to the patient's T cells, aiming to stimulate a robust and specific anti-tumor immune response. This strategy seeks to exploit the lymphopenic environment post-transplant to enhance the expansion and activity of tumor-specific lymphocytes.
