Overview

Tusamitamab ravtansine + pembrolizumab + cisplatin + pemetrexed is a quadruplet combination therapy being investigated for the first-line treatment of CEACAM5-positive non-squamous non-small cell lung cancer (NSQ NSCLC). The regimen combines tusamitamab ravtansine, an antibody-drug conjugate (ADC) targeting CEACAM5 with a maytansinoid payload (DM4), with the anti-PD-1 checkpoint inhibitor pembrolizumab and a standard chemotherapy doublet of cisplatin and pemetrexed. This approach aims to leverage the targeted cytotoxicity of the ADC, the immune-potentiating effects of the checkpoint inhibitor, and the systemic anti-proliferative effects of chemotherapy to improve outcomes in patients with advanced or metastatic disease. The combination is currently being evaluated in the Phase 2 CARMEN-LC05 clinical trial (NCT04524689).
