A combination regimen consisting of four agents: **tusamitamab ravtansine** (a CEACAM5-directed antibody-drug conjugate), **sintilimab** (an anti-PD-1 monoclonal antibody), **cisplatin** (a platinum-based chemotherapeutic), and **pemetrexed** (an antifolate chemotherapeutic). Tusamitamab ravtansine delivers a cytotoxic maytansinoid payload to cells expressing carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5), inhibiting microtubule assembly, while sintilimab blocks PD-1 signaling to augment antitumor immune responses. Cisplatin and pemetrexed interfere with DNA synthesis and function. This combination is being investigated in clinical trials primarily for advanced/metastatic non-squamous non-small cell lung cancer, particularly in tumors with high CEACAM5 expression[3][4][5][6].
