TV005 is a live attenuated tetravalent dengue vaccine developed by the National Institute of Allergy and Infectious Diseases (NIAID). It is designed to provide protection against all four serotypes of the dengue virus (DENV-1, DENV-2, DENV-3, and DENV-4). The vaccine is a mixture of four recombinant live attenuated dengue virus strains: rDEN1Δ30, rDEN2/4Δ30 (a chimeric strain), rDEN3Δ30/31, and rDEN4Δ30. Attenuation is achieved through specific deletions in the 3′ untranslated region of the viral genome and structural gene chimerization. It is typically administered as a single subcutaneous dose and has shown high levels of immunogenicity and safety in clinical trials, including Phase II trials in Bangladesh and Phase III trials in Brazil.
