Overview

TVB-2640 + chemotherapy refers to the combination of **TVB-2640**, a first-in-class, orally available, selective small molecule inhibitor of **fatty acid synthase (FASN)**, with various chemotherapeutic agents. TVB-2640 acts by **blocking de novo lipogenesis** (DNL) in tumor cells, thereby reducing steatosis and interfering with the metabolic support cancers need to grow and survive, especially under hypoxic conditions. It is under investigation for a range of solid tumors. In clinical trials, TVB-2640 has most frequently been combined with chemotherapies such as **bevacizumab** (an anti-VEGF monoclonal antibody) and **paclitaxel** (a taxane), as well as with other agents like **enzalutamide** (for prostate cancer) and **SN-38** (the active metabolite of irinotecan). The combination is in **phase I/II trials** for high-grade astrocytoma/glioblastoma, solid tumors, triple-negative breast cancer, and metastatic castration-resistant prostate cancer, among others. Developer and manufacturer have overlapping roles and include Sagimet Biosciences. TVB-2640 is not approved by the FDA or other major regulatory agencies for any indication as of late 2025[1][4][7][9].
