**U87 CAR-T + cyclophosphamide + interleukin-2** is an investigational combination therapy composed of U87-specific chimeric antigen receptor (CAR)-T cells, the cytotoxic agent cyclophosphamide, and the cytokine interleukin-2. U87 CAR-T cells are autologous T lymphocytes engineered to express a chimeric antigen receptor targeting a tumor-associated antigen present in the U87 glioblastoma/GBM cell line, commonly used as a model for head and neck and brain cancers[1][2]. Cyclophosphamide is a chemotherapeutic alkylating agent that may be used to deplete immune-suppressive cells and promote CAR-T cell expansion. Interleukin-2 is a cytokine that supports T cell growth, survival, and activation. The rationale for this combination is to enhance CAR-T cell anti-tumor activity, persistence, and tumor infiltration. This combination is in clinical investigation for the treatment of advanced head and neck tumors, including nasopharyngeal cancers[1].
