Drug intelligence / Profile preview
UB-VV500 + rapamycin is an investigational combination therapy consisting of UB-VV500, an in vivo chimeric antigen receptor (CAR) T-cell therapy, and rapamycin (sirolimus). Developed by Umoja Biopharma, UB-VV500 is built on the proprietary VivoVec lentiviral vector platform and is designed to target both B-cell maturation antigen (BCMA) and G protein-coupled receptor, family C, group 5, member D (GPRC5D) to address antigen escape and disease heterogeneity in multiple myeloma. The therapy incorporates a rapamycin-activated cytokine receptor (RACR) system. When rapamycin is administered, it selectively drives the expansion and persistence of the transduced CAR T-cells in vivo by converting rapamycin binding into an interleukin-2 (IL-2) and interleukin-15 (IL-15) signal, while simultaneously suppressing non-transduced immune cells, thereby eliminating the need for traditional lymphodepleting chemotherapy.
